What Are Enzymes?

Enzymes are proteins (and sometimes RNA molecules called ribozymes) that catalyze biochemical reactions. A catalyst is a substance that speeds up a reaction without undergoing permanent change.

  • Active Site: The region on the enzyme where the substrate binds and the reaction occurs.
  • Substrate: The molecule upon which the enzyme acts.
  • Product: The end result of the enzymatic reaction.

Enzymes are highly specific. The “lock and key model” explains that each enzyme fits only a particular substrate, similar to a key fitting a specific lock.


Functions of Enzymes

Enzymes perform an astonishing array of functions in the body:

  1. Digestive Enzymes: Break down large macromolecules into smaller, absorbable units.
    • Examples:
      • Amylase → breaks down starch into sugars
      • Lipase → breaks down fats into fatty acids
      • Protease → breaks down proteins into amino acids
  2. Metabolic Enzymes: Participate in energy production and metabolic pathways.
    • Example: ATP synthase helps produce ATP, the cell’s energy currency.
  3. DNA & RNA Processing: Enzymes like DNA polymerase and RNA polymerase are essential for replication and transcription.
  4. Detoxification Enzymes: Liver enzymes, such as cytochrome P450, detoxify drugs and harmful chemicals.

Factors Affecting Enzyme Activity

Enzymes are sensitive to environmental conditions. The main factors include:

  1. Temperature: Enzyme activity increases with temperature up to an optimum point. Beyond this, enzymes denature and lose function.
  2. pH: Each enzyme has an optimal pH. For example, pepsin works best in acidic conditions, while trypsin prefers basic conditions.
  3. Substrate Concentration: Increasing substrate concentration can increase the reaction rate until the enzyme becomes saturated.
  4. Enzyme Concentration: Higher enzyme concentration generally increases the reaction rate, provided substrates are available.

Types of Enzymes

Enzymes can be classified based on the reactions they catalyze:

  1. Oxidoreductases: Catalyze oxidation-reduction reactions.
    • Example: Lactate dehydrogenase
  2. Transferases: Transfer functional groups between molecules.
    • Example: Aminotransferases
  3. Hydrolases: Break bonds using water.
    • Example: Lipase, Amylase
  4. Lyases: Break bonds without water, forming double bonds.
    • Example: Decarboxylase
  5. Isomerases: Catalyze structural rearrangements within a molecule.
    • Example: Glucose-6-phosphate isomerase
  6. Ligases: Join molecules using ATP.
    • Example: DNA ligase
